Genotyping today. I took a few leaves from 12 plants, as well as Col-0. Liquid N2 and extraction using Sam’s DNA prep protocol. I am resuspending in 100ul TE. Using 1ul of template in each of two PCRs, JAK255/256 and JAK257/256.
They all had a single band the same size as the Col-0 wildtype. Need to figure out another way to check these, since perhaps one of the primer sets isn’t working.
